iSTEM是一款的小鼠胚胎干細胞維持培養(yǎng)基。該款產品包含三種小分子抑制物，從而阻斷干細胞分化的信號通路，同時維持細胞的多能干性（pluripotency）。 iSTEM可以被應用于難以獲得ES細胞的小鼠品系，獲取到ES細胞系(1)。

iSTEM培養(yǎng)基中的小鼠ES細胞與在添加了Leukemia Inhibitory Factor (LIF) 和 serum/Bone Morphogenetic Protein (BMP)的培養(yǎng)基的細胞相比，增殖速度加倍(1)。另外，在iSTEM中培養(yǎng)的小鼠ES細胞產生細胞聚落（clone）的能力顯著提高（enhanced clonogenicity）,從而可以在單細胞鋪板后（single cell deposition），產生更多的干細胞克隆。

iSTEM is a compley defined cell culture medium for deriving, maintaining, and propagating mouse embryonic stem (ES) cells. This media formulation captures mouse ES cells in their basal state by blocking the inductive pathways of differentiation with selective small molecule inhibitors. The medium contains three small molecule inhibitors that block differentiation-inducing signals and promote cell survival, enabling maintenance of pluripotency without the need for additional stimulatory cytokines. iSTEM medium can also be used to derive mouse ES lines from strains that have proven refractory to ES cell derivation under standard conditions (1).

Mouse ES cells cultured in iSTEM medium continually expand at a doubling rate that is comparable to that of cells cultured in the presence of Leukemia Inhibitory Factor (LIF) and serum/Bone Morphogenetic Protein (BMP) (1). In addition, culture in iSTEM medium enhances clonogenicity; a higher frequency of pluripotent colonies are established after single cell deposition.

自備試劑：

For mouse ES cells

Use culture vessels precoated with 0.1% gelatin type A in PBS (at room temperature for 15 minutes or more), or precoated sequentially with 0.01%poly-L-ornithine hydrobromide (at 37℃ for 30 minutes or more), washing twice with PBS, followed by 10 μg/mL laminin in PBS (at 37℃ for 3 hours or more).

對于小鼠ES細胞

使用在PBS中預涂有0.1％明膠A型的培養(yǎng)容器（在室溫下15分鐘或更長時間），或者依次用0.01％聚-L-鳥氨酸氫溴酸鹽（在37℃下30分鐘或更多）預涂覆，用PBS洗滌兩次 ，然后用PBS中的10μg/ mL層粘連蛋白（37℃下3小時以上）。

保存：Upon receipt, store at -20℃ until ready to use. When stored under these conditions,the product is stable for 12 months from the date of manufacture (see label).Once thawed, store at 4℃ and use within 2 weeks.This product is light sensitive, and should be protected from light.
